Resin especially designed for the synthesis of long and difficult peptide sequences!Due to modifications of the polystyrene backbone, TentaGel XV shows extended swelling properties providing a large reaction volume. It is a superior resin for synthesizing difficult sequences, aggregating peptides, mini proteins, and PNAs in high purity and excellent yields. The dedicated crosslinking design in TentaGel XV leads to increased swelling, which is similar or even higher compared to PEGA® or ChemMatrix® resins. The polar PEG spacers allow the composite resin to swell in a wide range of solvents including water, methanol, DMF, DCM and make its handling very easy.
The high swelling creates an extended reaction space providing ideal conditions for the synthesis of difficult sequences, aggregating peptides, mini proteins and PNAs in high purities and excellent yields.

DTNB, frequently called Ellman’s Reagent, used as reagent for determination of free sulfhydryl groups. Through reaction with aliphatic thiol groups a mixed disulfide of protein thiol and one mole of 2-nitro-5-thiobenzoate per mole of protein sulfhydryl group is being formed. DTNB has little absorbance. Reaction with -SH groups on proteins (from any solvent accessible Cys) under mild alkaline conditions (pH 7-8) produces the 2-nitro-5-thiobenzoate anion, which gives an intense yellow color with an absorption maximum at 409.5 nm (Extinction coefficient: 14150 M -1*cm -1). Ellman’s reagent is sensitive to various buffer ions, therefore, the extinction coefficient used to calculate the number of sulfhydryl groups must be matched to the reaction conditions. In case the thiol groups are in disulfide bonds, they must be reduced under anaerobic conditions prior to reaction with DTNB.
Solubility 8 mg/mL in EtOH. Stock solutions c an be prepared in 0.1 M phosphate buffer, which are stable over several weeks.

In nature, the American firefly (photinus pyralis) uses the luciferase bioluminescence to find a partner for reproduction. The result of the luciferase reaction is the emission of yellow-green light at 560 nm. This reaction requires the enzyme luciferase, the substrate D-Luciferin and the co-factors ATP, Mg2+ and Oxygen. The reaction is a cascade of several steps: the initial adenylation of Luciferin is followed by oxidation to Oxyluciferin. The Oxyluciferin resulting from this reaction is in an electronically excited state. On relaxation to ground state a photon is released (from 550 nm (yellow-green) at neutral pH to 620 nm (red) at low pH). In modern biotechnology there are numerous applications for the Luciferase system. |

PGA (Penicillin G Amidase, Penicillin Acylase, Penicillin Amidohydrolase from E.coli on acrylic resin, Systematic name: Penicillin amidohydrolase, E.C. 3.5.1.11) has an active pocket, which is very specific for phenyl acetic acid. The prominent commercial use is hydrolysis of a phenylacetamid bond during production of the penicillin API 6-APA (De Martin et al., J. Mol. Catal. B:Enzymatic 1999; 6: 437). The high specifity of PGA towards the Phenylacetyl moiety makes the use of Phacm as new alternative for Acm very promising. The principle use already was discovered by Albericio et al in 1995 using native PGA. Today PGA is available as immobilized preparation, so by simple filtration process PGA can be removed from the reaction mixture and reused many times.

For the modification of Gold surfaces sulphur bearing dendrons can be used, they also conjugate with biologicals via disulfide bridge formation.

After cleavage from the resin (with trifluoroacetate using triethylsilane or ethanedithiol as scavenger) a peptide is being designed, which carries a thiol group on the C-terminus. As sulphur has high affinity to heavy metals, like Zn, Fe or Ni, which are essential metal centers in metallo proteins, the affinity of peptide substrates or pharmacological active sequences can further be increased through a strong sulphur-metal interaction.
