PEGylation

PEGylation

PEG derivatives, the pioneers for drug delivery, are available from pure, monodisperse, discrete molecules with short chain lengths to long polydisperse, allowing for regio-specific chemical conjugation with small molecules, proteins, peptides, and biopharmaceuticals through their broad variety of terminal chemical groups available. They can be attached to different positions – most prominent candidates are amino groups of solvent accessible lysines, N-termini, or thiol functions of available cysteines. Rarely used is the C-terminus or carboxylic groups from aspartate and glutamate. Besides, PEG can also serve as spacer or cross linker between two moieties.
